Abstract

The influence of intracellular calcium dynamics on spontaneous activity of pacemaker cells was investigated via mathematical modeling. We compared the responses to the inhibition of calcium L-type current in several models of electrical activity of sinoatrial node cells. All models showed a decrease in the maximum velocity of depolarization, action potential amplitude, action potential duration. Calcium clock models showed an increase in the period of calcium oscillation up to 12%. Models in which spontaneous activity is mainly determined by the hyperpolarization-activated current showed up to 15% reduction in the oscillation period. A comparison of theoretical results with experimental data indicates different contribution of intracellular mechanisms to spontaneous activity of pacemaker cells from center and periphery of the sinoatrial node.
